Ejaculate Motility

The motility of the sperm in an ejaculate is important to determine because it will be a factor in detemining the dose of a fresh ejaculate or whether to ship or freeze an ejaculate. There are machines which measure the sperm concentration and motility, but they are very expensive (>$33,000 + microscope). A good quality microscope (optional camera output to a monitor) is all that is needed. The exact % of sperm that are motile is not critical so much as an estimate of the motility. One procedure to determine motility is shown below.

Things that effect motility:

  • Temperature
  • Time after dilution into extender
  • Substrates (ie., sugars, caffeine)
  • Extenders
  • Urine or water in ejaculate will severely decrease motility
  • Time since last collection
  • Frequency of collection
