Fresh Semen Preparation for Insemination
Initial Steps:
- Remove 1 ml raw semen
- Evaluate semen for concentration, motility and record volume in ml
- Dilute remaining semen 1:1 with warm extender, place on counter.
- Calculate motile sperm /ml in the extended semen (MS/ml)
- MS/ml = (concentration/2)* % motility
Insemination Volume (Inseminate
500 X 10^6 motile sperm in a volume of at least 10 ml):
- Insemination Volume = 500 X 10^6 / (MS/ml)
- Units will be in ml
- If <10 ml then added sufficient extender to insemination
dose to bring to 10 ml
- If >10 ml just inseminate volume determined.
Assumptions:
- Hold extended semen at around 21 – 22°C
- place in thermos to ensure semen is not cold shocked
- Insemination to occur within 1 – 2 hours
- If semen will be held 2 – 4 hours, dilute semen
at least 1:2 or 1:3
- For longer holding times cool semen following appropriate
procedures
