90% Percoll

1. Make a stock solution (Percoll Salts) with the following composition:
   
   
   

   Chemical	gm/100 ml
   KCl	0.230
   NaH2PO4.H2O	0.040
   NaCl	4.080
   Hepes	9.530
   Adjust to pH 7.4 (room temp)

   
   Filter to sterilize (0.22 micron filter) and store the Percoll Salts at 5 C. Do not mix CaCl₂, MgCl₂ or NaHCO₃.
   
   
2. To make the 90% Percoll solution, add 45 ml Percoll, 5 ml of the Percoll Salts Solution and the following components with stirring:
   
   
   

   Chemical	amount/50 ml
   CaCl2.2H2O	14.7 mg
   MgCl2.6H2O	4.0 mg
   Lactic Acid (Sodium salt; 60% w/w syrup)	0.184 ml
   NaHCO3	42 mg

   
   
   Filter to sterilize (0.22 micron filter) and store at 5 C.
